Lodenafil carbonate is a PDE5 inhibitor developed and marketed by Cristália in Brazil under the brand name Helleva.
Two molecules in one
Lodenafil carbonate is made of two lodenafil molecules joined together. The body splits it into two molecules of the active drug, a design meant to improve absorption. In laboratory tests lodenafil was roughly twice as potent as sildenafil at blocking the PDE5 enzyme.
Evidence
A Brazilian phase III trial randomized 350 men with ED to placebo, 40 mg or 80 mg. Both doses improved erectile function scores compared with placebo. Reported side effects included headache, flushing, rhinitis and indigestion, the same pattern as other PDE5 inhibitors.
Approval
Lodenafil is used in Brazil. It is not authorized in the EU, UK or US.
